The Canon EOS M100 is designed to block the shutter release without a lens to protect the sensor and internal mechanisms. However, contrary to what the old answer suggests, Canon offers an official custom function to allow this practice if needed.

Activation via custom function C.Fn III-4: Access the Customization menu, then select [C.Fn III:Operation/Others]. Look for the custom function C.Fn III-4: Trigger shutter without lens. This function has two options: 0: Off (default setting, blocks shutter release) and 1: On (allows shutter release without a lens). Select 1: On to enable this feature.

Procedure to access the menu: Press the Menu button, go to the Customization tab (key icon), then navigate to [C.Fn III:Operation/Others]. Use the navigation buttons to select the custom function number (III-4), then change the setting with the W (previous) and X (next) buttons to switch to 1: On. Press M to confirm and exit.

Warnings and precautions: Once this function is activated, ensure that the camera is clean and that access to the sensor is safe before triggering without a lens. The 24.1 MP CMOS sensor remains exposed to dust and elements. This option is useful for digiscoping (astronomical observation with the camera), using specialized adapters, or testing, but disable it as soon as possible after use.

Deactivation: To restore the default protection, return to C.Fn III-4 and set the parameter to 0: Off. This will prevent any accidental shutter release without a lens.
